Second-round matches were played at the Masters in Cincinnati.

At this stage, fifth-seeded Kasper Ruud lost to Max Purcell of Australia and Holger Rune was unable to finish the match against Mackenzie McDonald. Emil Ruusuvuori stopped the seventh seeded Andrey Rublev.

After winning the Masters in Toronto, Yannick Sinner lost early in Cincinnati and was stopped by Dusan Lajovic.
